Competent Cell의 종류는 정말 많습니다.
E.coli가 가지는 유전자형에 따라서, 용도에 따라서, Transformation 방법에 따라서 나뉘어지죠. 여기서는 가장 보편적으로 이용하는 competent E. coli strain의 종류 및 특성에 대해서 간단히 비교 분석을 하겠습니다.
③ XL1-Blue competent E.coli
* 재조합 plasmid를 위한 blue-white color screening에 이용하며 또한 일반적인 plasmid 또는 lamda vector로 cloning할 때 사용한다.
* Genotype of E.coli strain XL1-Blue
*- tetracycline resistant
*- endonuclease(endA)deficient -> DNA miniprep시 질적인 향상.
*- recombinantion (recA)deficient -> Insert DNA의 안정성을 증가
*- LsdR mutation -> EcoK endonuclease에 의한 cloned DNA가 잘리는 것을 방지.
* IPTG and X-gal color screening 가능 (F’ episome 안에 lacIqZΔM15 gene으로 인해 가능)
* 통상적으로 45초가 heat shock을 시행한다.
* 재조합 plasmid를 위한 blue-white color screening에 이용하며 또한 일반적인 plasmid 또는 lamda vector로 cloning할 때 사용한다.
